Yesung officially debuted with the 12-member project group Super Junior 05 on November 6, 2005 on the SBS music program Inkigayo, performing their first single, "Twins (Knock Out)". Their debut album SuperJunior05 (Twins) was released one month later on December 5, 2005 and debuted at # 3 on the MIAK K-pop weekly album chart.

From September 2006 to September 2007, Yesung hosted a radio program, M.I.R.A.C.L.E for You, in which Super Junior members often appeared as guest stars. However, before the radio's first anniversary, Yesung left to prepare for Super Junior's second album, Don't Don. This album became the second best-selling album in South Korea. This radio show last aired on September 8, 2007.

In November 2006, Yesung, along with fellow group Ryeowook and Kyuhyun, joined Super Junior's first sub-group, Super Junior-K.R.Y., A group that focuses on singing R&B ballads. The trio debuted on November 5, 2006 on the KBS music show Music Bank with the soundtrack for the drama Hyena entitled "The One I Love".
