《梦溪笔谈》包括《笔谈》、《补笔谈》、《续笔谈》三部分，收录了沈括一生的所见所闻和见解。《笔谈》二十六卷，分为十七门，各卷依次为“故事（一、二）、辩证（一、二）、乐律（一、二）、象数（一、二）、人事（一、二）、官政（一、二）、机智、艺文（一、二、三）、书画、技艺、器用、神奇、异事、谬误、讥谑、杂志（一、二、三）、药议”。《补笔谈》三卷，包括上述内容中十一门。《续笔谈》一卷，不分门。全书共六百零九条（不同版本稍有出入），内容涉及天文、历法、气象、地质、地理、物理、化学、生物、农业、水利、建筑、医药、历史、文学、艺术、人事、军事、法律等诸多领域。在这些条目中，属于人文科学例如人类学、考古学、语言学、音乐等方面的，约占全部条目的18%；属于自然科学方面的，约占总数的36%，其余的则为人事资料、军事、法律及杂闻轶事等约占全书的46%。
从内容上说，《梦溪笔谈》以多于三分之一的篇幅记述并阐发自然科学知识，这在笔记类著述中是少见的。如《技艺》正确而详细记载了“布衣毕升”发明的泥活字印刷术，这是世界上最早的关于活字印刷的可靠史料，深受国际文化史界重视。“辩证”门谈韩愈画像条，使后人了解从北宋就产生并沿袭下来的一个错误：把五代韩熙载的写真当成韩愈的画像。
