Prof. Dr. KH. Ali Mustafa Yaqub, MA lahir di Kemiri, Batang, Jawa Tengah, pada tanggal 2 Maret 1952 M dari sebuah keluarga yang taat menjalankan agama.Domisili beliau sekarang di Jl. SD. Inpres No. 11 RT.002 RW.09 Pisangan-Barat Ciputat 15419 Tangerang-Selatan Banten.
Pendidikan KH. Ali Mustafa Yaqub mulai dari SD sampai SMP, semua dijalani di Batang kota kelahirannya. Setelah tamat SMP minatnya untuk belajar agama mulai tumbuh, Ali Mustafa kecil bertandang ke sebuah pesantren di Seblak, Jombang untuk belajar agama sampai tahun 1969.
Kemudian beliau nyantri lagi di pesantren Tebu Ireng, Jombang sampai tingkat Fakultas Syari’ah Universitas Hasyim As’ari sampai awal tahun 1976. Dan pada tahun itu juga ia masuk Fakultas Syari’ah Universitas Muhammad ibnu Saud sampai tahun 1985 kemudian mengambil Master di Universitas yang sama pada Jurusan Tafsir dan Ilmu Hadits.
